Turn on: first turn on the switch on the other unit, then turn on the switch on the back of the main unit.
Shutdown: first turn the parameter status of all units on the controller to off, turn off the air intake switch and then shut down; when shutting down, turn off the switch behind the controller, and then turn off the switches on other units.
Sterilization operation
1. Remove the motor and lay it flat on the table. Put a black protective cap on the top of the can.
2. Remove the temperature sensor from the tank. The sensor does not need to be sterilized.
3. Remove the PH cable and put on a red protective cap; insert the PH electrode to the end. Be sure to tighten the upper and lower fixing nuts of the electrode. The PH electrode should be calibrated before sterilization. PH electrode calibration: After connecting the cable, unscrew the fixing nut of the electrode (the first one), take out the PH electrode, rinse it with deionized water, gently absorb the water with tissue paper and put the standard solution first Calibrate in 4.01. After the data on the controller is stable, enter 4.01 at set zero (regardless of the slight difference of the actual value); after the input is completed, remove the electrode from the standard solution, rinse it, absorb it and place it in the 7.00 standard solution After the data on the controller is stable, enter 7.00 at the set span (regardless of the slight difference in actual values). Do not turn off the controller after PH electrode calibration.
4. Remove the DO cable, wrap the DO cable interface with tin foil and put on a black cap. Calibration after DO electrode sterilization. To insert the DO electrode to the end, tighten the upper and lower fixing nuts of the electrode. Before sterilization, pay attention to whether there is any protective solution in the dissolved oxygen electrode, and if not, add it.
5. Remove the defoaming or liquid level cable, insert the defoaming or liquid level cable to the end, do not press down after positioning to the required height after sterilization. The height of the defoaming electrode is about 1.5 cm away from the liquid surface.
6. Remove the inlet and outlet water pipes on the fermentation tank and the condenser. At the same time, remove the joints of the removed water pipes with clips to prevent water from flowing out.
7. Both ends of the filter on the fermentation tank should be clamped with clips, the outlet of the filter should be wrapped with tin foil, and the ends of all filters should be clamped with clips. After the fermentation is completed, pour the tank and check whether the filter is wet. If so, blow out the liquid with a suction ball as soon as possible.
8. Clamp the silicone tube on the harvest tube with an iron clip to prevent running liquid in the sterilization pot. When the harvest tube is not in use, it must be stuck and not removed.
9. Before sterilization. Unscrew the six fixed nuts of the fermentor to let out the gas, other nuts must be tightened! After sterilization, open the pressure cooker and tighten it immediately. Take care to wear gloves to prevent scalding.
10. Do not put the acid in the autoclave to sterilize. Wrap the filter on the empty acid bottle with tin foil and unscrew the cap to release steam. After sterilization, remove the foil paper on the foil filter as a steam outlet. (Note that the silicone tube connected to the acid bottle should not be stuck); the lye bottle and the silicone tube can be sterilized together (the silicone tube connected to the lye bottle should be stuck).
11. Remove the electric heating jacket and tile it on the desktop.
12. Adjust the position of the stirring blade before sterilization. The bottom stirring blade should be as close as possible to the air distributor, and the upper position should be adjusted to the middle position of the added fermentation broth.
13. The position of the sampler should not be close to the position of the baffle, as close to the middle position as possible.
14. Remove the suction ball of the sampler before sterilization. Do not lean all silicone tubes against the inner wall of the pressure cooker.
After sterilization
1. Tighten the six loosening screw caps and acid-base caps.
2. Remove the clips of tinfoil and silicone tube wrapped in the filter.
3. Install the motor. If the lubricating oil inside the motor is dry, apply an appropriate amount of petroleum jelly.
4. Calibrate the DO electrode after connecting the dissolved oxygen cable: After the sterilization, connect the DO electrode cable and enter 0 at set zero; control the air flow of the reactor at 1L / min and Agitation to 100r / Min overnight, open in the morning, enter 100 in the set span and press the enter key.
5. Connect the inlet and outlet pipes, PH cables and defoaming cables when the temperature drops to the temperature required for fermentation.
6. Before putting in the temperature electrode, add glycerin dropwise to the inlet of the temperature electrode. After inserting the electrode, continue adding dropwise until it comes out. Note that the temperature electrode should be inserted all the way.
7. Check the output pressure of the air compressor, it should be in the range of 0.7 ~ 1.0kg / cm3.
8. The silicone tube of defoamer should be installed on the pump in order from bottom to top.
Set the working temperature of the chiller. Turn on: Connect the inlet and outlet pipes → turn on the main power supply → carry out temperature design → turn on the circulation pump → start refrigeration and shutdown: first turn off the refrigeration switch → turn off the circulation pump switch → turn off the inlet and outlet valves at the same time → turn off Power temperature setting: turn on → the panel displays "PP0.0" → press "shift key" and "adjustment key" and display "PP0.1"

Prepare the inoculation preparation after the above operation: prepare iron baffle, lighter, alcohol spray can, inoculation ring, 95% ethanol enamel cylinder container, pointed pliers, adjustable wrench, inoculation bottle.
Vaccination operation
1. Remove the hose near the inoculation port, surround the inoculation port with an iron baffle, and spray alcohol at the inoculation port with an alcohol spray can. (Note: If a flame overflows the iron plate along with alcohol, it needs to be blown out Flames overflowing on both sides of the iron plate.)
2. Dip the inoculation ring with alcohol and place it on the inoculation port. After dipping the wrench or pliers with alcohol, unscrew the nut and clamp it out with the pliers. Blow out the flame on the nut and pliers and put it in In a 95% ethanol enamel cylinder container.
3. After unplugging the inoculation bottle at the flame, quickly pour it into the fermentation tank. After finishing, clamp the nut with pliers and screw on. The entire process ensures aseptic operation!
Dissolved oxygen setting: There are four conditions for the control of dissolved oxygen in fermentation mode: press off key → press control key → press Auto key → setup → cascade → select the following four modes.
1. None state: The control of dissolved oxygen is mainly controlled by increasing the air flow and setting a fixed speed.
2. Agit state: The control of dissolved oxygen is mainly controlled by increasing the rotation speed. In this state, when the air flow rate remains unchanged, the dissolved oxygen is increased by increasing the rotation speed when the cell consumption decreases. Until the speed reaches the set upper speed limit. This process is automatically controlled.
3. 02 state: This state is mainly controlled by controlling the mixing percentage of oxygen and air. Dissolved oxygen increases as the percentage of oxygen increases. For automatic control process.
4. Agit / 02 state: In this state, the control of dissolved oxygen is first increased by increasing the speed. When the speed reaches the set upper limit, if the actual dissolved oxygen value still does not reach the required setting value, the speed Control at the upper limit, and start to automatically increase the oxygen intake in a small to large percentage until it reaches the set dissolved oxygen value. This process is also an automatic control process.
Note: When using the Agit state and Agit / 02 state, if the actual dissolved oxygen value is too different from the set value, make sure that the upper limit of the set stirring speed is not too large, otherwise the speed will increase within a moment. Too much shear force and a lot of foam. In actual operation, it can be assisted by manual control of the air flow meter.
